No, on WordPress.com, you can only assign passwords to posts or pages, not uploaded files.

You can have secured PDF documents on WordPress.COM –
I put a password on them to prevent the file from being changed – but I let them be printed –
never tried putting a password on one for read access but I assume that would also work – you do need to put the password on the PDF document when you create it on your PC –

That would certainly work, though it doesn’t relate directly to WordPress.com. I read the OP’s question to mean if they could password protect the download, not the document itself.
Paid Box.com accounts allow you to password protect any download, but it’s a steep price (15$ a month) to pay for just that option and makes me wonder if there aren’t other alternatives as well.
