<title><![CDATA[Brain Oxytocin: A Key Regulator of Emotional and Social Behaviours in Both Females and Males]]></title>

<content:encoded><![CDATA[<p align="justify"><font size="3"><img alt="Brain and Body_300" hspace="15" src="http://hypericum.files.wordpress.com/2008/06/brain-20and-20body-300.jpg" align="right" vspace="5" border="0" />In addition to various reproductive stimuli, the neuropeptide </font><a href="http://www.oxy-tocin.com/index.php?lang=1"><font size="3">oxytocin</font></a><font size="3"> (OXT) is released both from the neurohypophysial terminal into the blood stream and within distinct brain regions in response to stressful or social stimuli. Brain OXT receptor-mediated actions were shown to be significantly involved in the regulation of a variety of behaviours. Here, complementary methodological approaches are discussed which were utilised to reveal, for example, anxiolytic and anti-stress effects of OXT, both in females and in males, effects that were localised within the central amygdala and the hypothalamic paraventricular nucleus. Also, in male rats, activation of the brain OXT system is essential for the regulation of sexual behaviour, and increased OXT system activity during mating is directly linked to an attenuated anxiety-related behaviour. Moreover, in late pregnancy and during lactation, central OXT is involved in the establishment and fine-tuned maintenance of maternal care and maternal aggression. In monogamous prairie voles, brain OXT is important for mating-induced pair bonding, especially in females. Another example of behavioural actions of intracerebral OXT is the promotion of social memory processes and recognition of con-specifics, as revealed in rats, mice, sheep and voles. Experimental evidence suggests that, in humans, brain OXT exerts similar behavioural effects. Thus, the brain OXT system seems to be a potential target for the development of therapeutics to treat anxiety- and depression-related diseases or abnormal social behaviours including autism.</font> </p>

<title><![CDATA[Industry junks St John's wort ADHD study]]></title>

<content:encoded><![CDATA[<p align="justify"><font size="3"><img alt="Adhd1" hspace="10" src="http://hypericum.files.wordpress.com/2008/06/adhd1.jpg" align="left" vspace="10" border="0" />A study that found St John's wort was ineffective in positively affecting children suffering from attention-deficit hyperactivity disorder (ADHD) has been heavily criticized by industry for being badly designed. Industry has criticized the study and disputed the researchers claim that St John's wort is one of the most common treatments for ADHD among children.</font></p>
<p align="justify"><font size="3">It has also been criticized for subjecting St John's wort to inappropriate, pharmaceutical-style testing when the herb also known as <em>Hypericum perforatum</em> does not function as a drug.<em>"Why wasn't a gold standard [double blind, placebo-controlled] used to do a proper comparison?"</em> Daniel Fabricant, PhD, vice president of scientific and regulatory affairs at the Natural Products Association (NPA) told NutraIngredients-USA.com.</font></p>
<p align="justify"><em><font size="3">"If they're going to set up a drug trial then they should go all out with it. We all know St. John's wort isn't a drug, and traditionally isn't used&#160;or intended to be used as a drug."</font></em></p>
<p align="justify"><font size="3">St John's wort's most commonly studied benefit is in the relief of mild-to-moderate depression and anxiety.</font></p>
<p align="justify"><font size="3"><em>"It's commonly used and has shown&#160;efficacy for mild-to-moderate depression in more appropriately designed trials,"</em> Fabricant said.&#160;<em>"But if you are going to treat it like&#160;a drug&#160;in a trial, then all the bells and whistles have to be there to accurately tell the whole story. In this instance if the gold standard intervention showed the same lack of effect, would it have been reported that the gold standard didn't work for ADHD?"</em></font></p>
<p align="justify"><font size="3">He added: <em>"It certainly would have nailed home the lack of quality in this study and made it significantly more difficult to publish and call it peer-reviewed academic medical science."</em></font></p>
<p align="justify"><strong><font size="3">Usage and quality</font></strong></p>
<p align="justify"><font size="3">The Mayland-based American Herbal Products Association (AHPA) questioned the quality of the St John's wort used in the trial and disputed the researcher's assertion that St John's wort is commonly used to treat ADHD among young people.</font></p>
<p align="justify"><font size="3"><em>I don't believe that this test material would pass muster if the proposal was submitted to NIH's National Center for Complementary and Alternative Medicine (NCCAM) today because something as basic as ensuring the quality through the duration of the trial was not assured,"</em> said Steven Dentali, PhD, AHPA's chief scienceofficer and past chairperson of NCCAM's Product Quality Working Group.</font></p>
<p align="justify"><font size="3">AHPA president, Michael McGuffin, said he knew of no manufacturers marketing St John's wort at children.</font></p>
<p align="justify"><em><font size="3">"It's an out-of-the-ordinary, rare use. It might be a use that some mom thinks makes sense," he said.</font></em></p>
<p align="justify"><font size="3">The randomized and placebo-controlled trial was published on June 11 in the <em>Journal of the American Medical Association</em> and involved 54 children ages 6 to 17 years.</font></p>
<p align="justify"><font size="3">Half were given St John's wort, while the other half were given a placebo and no significant difference was found between the two groups in improving the symptoms of ADHD.</font></p>

After 5 years of lying, it felt good! Great actually. My parents, (life long Republicans!) were pretty good about it too. Apparently they suspected I was gay, not so much because I acted gay, I don't really, but because all I ever brought home were handsome buff guys from soccer practice, or track meets, never ever girls. Conservative but loving parents they waited (against all hope) that they were wrong rather than confront me. My stupid old Grandfather was a whole other story but I won't go into that!</span></div>

<div><span style="font-size:small;">I also came out because I couldn't handle the constant lying. At the end of the day, like my parents, I'm a fairly conservative guy; monogamous by nature and all of the lying was decaying my personal code of ethics. Furthermore, all of the compartmentalization necessary to maintain the lies was a lot of work. I also thought coming out would give me a better chance of finding landing a partner. One of my favorite pastimes in trigonometry class was daydreaming about Mr. Right. </span></div>

<div><span style="font-size:small;">As your average garden variety gay guy will tell you, finding Mr. Right in the "gay world" isn't so easy. Oh, it's real easy if you like the flakey, lispy, effeminate types, but those people turn me off, big time. I gravitate to the straighter types. My father always used to say that water seeks it's own level so maybe it's just the potential of camaraderie that the straighter guys sensed? It could also be that I'm shorter and slighter than average, which I may be attractive to alpha type guys? Whatever. </span></div>

<div><span style="font-size:small;">I eventually found a great guy and entered into a monogamous relationship that lasted about three years. Then one day Bill ended it by reason of, get this, I was getting too muscular and I threatened his masculinity. He said, "Kittens grow into cats." Irony of ironies, he was the Tom cat, walking the fence line in every alleyway in town. I'm not the brightest bulb on the tree but that excuse was too simplistic, too pat. <title><![CDATA[Oxytocin and cortisol in romantically unattached young adults: Associations with bonding and psychological distress]]></title>

Cortisol was related to attachment anxiety. Regression analysis indicated that the adult's representations of bonding to parents predicted OT levels above and beyond cortisol, psychological distress, and attachment. The peptide affects steroidogenesis and sperm transport in vivo. Effects of OT on sperm motility in vitro seems to be contradictory. As no data are available on the presence of OT in human semen and on the relationship of OT with sperm characteristics, we assessed OT level in semen samples in 3 groups of patients: (I) normozoospermic, (II) astheno-/oligo-/teratozoospermic, and (III) azoospermic subjects. Furthermore, we studied the relationship between the concentration of OT in semen and the sperm characteristics. OT was measured in seminal plasma by radioimmunoassay after extraction. OT semen levels did not differ in control patients (I: 1.72 +/- 0.78 pg/mL; n = 10), patients with poor semen quality (II: 1.66 +/- 0.91 pg/mL; n = 11), or in vasectomized patients (III: 1.28 +/- 0.65 pg/mL; n = 11). No statistically significant relationships between the OT levels and sperm characteristics (density: 0.0693; total sperm count: 0.0845; percentage of motility: 0.1341; morphology: 0.3478) have been found. The neurosecretory peptide </font><a href="http://www.oxy-tocin.com/" target="_blank"><font size="3">oxytocin</font></a><font size="3"> is present in human seminal plasma of normal as well as of vasectomized subjects. OT is not only derived from the testis; OT levels in poor semen samples are not different from controls. No relationship was found between OT seminal plasma levels and sperm characteristics.</font></p>

Medical conditions, effect of substances, and psychological issues were ruled out. The patient was properly consented to using oxytocin as an off-label trial. Oxytocin was administered using a nasal spray intracoitally because of its ultra-short half-life. Results. </font></div>

However, sites of action and the intracellular signalling pathways downstream of OT receptors (OTR) within the brain remain largely unknown. In the present studies, we localized the anxiolytic effect of OT by bilateral microinfusion of OT (0.01?nmol/0.5?&#181;L) into the hypothalamic paraventricular nucleus (PVN) in male rats using both the elevated plus-maze and the light-dark box. Moreover, intracerebroventricular administration of OT, but not of the related neuropeptide vasopressin (VP), dose-dependently activated the extracellular signal-regulated kinase 1/2 (ERK1/2) cascade. Specifically, OT induced the phosphorylation of Raf-1, MEK1/2 and ERK1/2 in the hypothalamus <i>in vivo</i> and in hypothalamic H32 neurons via EGF receptors. OT-induced ERK1/2 phosphorylation was immunohistochemically localized within VP neurons of the PVN and the supraoptic nucleus. Importantly, the anxiolytic effect of OT within the PVN was prevented by local inhibition of the MAP kinase cascade with a MEK1/2 inhibitor (U0126, 0.5?nmol/0.5?&#181;L) locally infused prior to OT, indicating the causal involvement of this intracellular signalling cascade in the behavioural effect of OT. OT effects within the hypothalamus may have far-reaching implications for the regulation of emotionality and social behaviours and, consequently, for the development of possible therapeutic strategies to treat affective disorders. Although OT-like substances have been identified in all vertebrates, OT has been found only in mammals where it plays a major role in the onset and maintaining of behaviors which are typical of these animals, such as labour and lactation. Recently, several data have suggested the involvement of OT in the formation of infant attachment, maternal behavior, pair bonding and, more generally, in linking social signals with cognition, behaviors and reward. </p>
<p align="justify"><font size="3"></font></p>
<p align="justify"><font size="3">The aim of this paper was to review critically the role of OT in the regulation of different physiological functions and complex behaviors, as well as its possible involvement in the pathophysiology of some neuropsychiatric disorders. MEDLINE and PubMed (1972-2007) databases were searched for English language articles by using the following keywords: </font><a href="http://www.oxy-tocin.com/" target="_blank"><font size="3">oxytocin</font></a><font size="3">, physiology, cognitive functions, attachment, psychopathology, psychiatric disorders. Papers were examined that addressed the following aspects of the OT system: synthesis and localization, receptors, physiology: In addition, latest findings showing abnormalities of OT and OT system in several neuropsychiatric disorders, including autism, obsessive-compulsive disorder, eating disorders, addiction, schizophrenia, post-traumatic stress disorder and Prader-Willy syndrome, will be also discussed together with the possible clinical use of OT or its analogues and/or antagonists.</font></p>

<p align="justify"><font size="3"><img alt="Injection_300" hspace="10" src="http://hypericum.files.wordpress.com/2008/06/injection-300.jpg" align="right" vspace="10" border="0" />The neuropeptide </font><a href="http://www.oxy-tocin.com/" target="_blank"><font size="3">oxytocin</font></a><font size="3"> (20-100?ng), induces penile erection when injected unilaterally into the caudal but not rostral mesencephalic ventral tegmental area (VTA) of male Sprague-Dawley rats. Such pro-erectile effect started 30?min after treatment and was abolished by the prior injection of d(CH<sub>2</sub>)<sub>5</sub>Tyr(Me)<sup>2</sup>-Orn<sup>8</sup>-vasotocin (1?&#181;g), an oxytocin receptor antagonist injected into the same caudal ventral tegmental area or of haloperidol (1?&#181;g), a dopamine receptor antagonist, injected either into the nucleus accumbens shell (NAs) or into the paraventricular nucleus of the hypothalamus (PVN) ipsilateral to the injected ventral tegmental area. Penile erection was seen 15?min after the occurrence of, or concomitantly to, an increase in extracellular dopamine and its metabolite 3,4-dihydroxyphenylacetic acid (DOPAC) in the dialysate obtained from the nucleus accumbens or the paraventricular nucleus, which was also abolished by d(CH<sub>2</sub>)<sub>5</sub>Tyr(Me)<sup>2</sup>-Orn<sup>8</sup>-vasotocin (1?&#181;g), injected into the ventral tegmental area before </font><a href="http://www.oxy-tocin.com/" target="_blank"><font size="3">oxytocin</font></a><font size="3">. In the caudal ventral tegmental area oxytocin-containing axons/fibres (originating from the paraventricular nucleus) appeared to closely contact cell bodies of mesolimbic dopaminergic neurons retrogradely labelled with Fluorogold injected into the nucleus accumbens shell, suggesting that oxytocin effects are mediated by the activation of mesolimbic dopaminergic neurons, followed in turn by that of incerto-hypothalamic dopaminergic neurons impinging on oxytocinergic neurons mediating penile erection. As the stimulation of paraventricular dopamine receptors not only induces penile erection, but also increases mesolimbic dopamine neurotransmission by activating oxytocinergic neurons, these results provide further support for the existence of a neural circuit in which dopamine and </font><a href="http://www.oxy-tocin.com/" target="_blank"><font size="3">oxytocin</font></a><font size="3"> influence both the consummatory and motivational/rewarding aspects of sexual behaviour.</font></p>

<content:encoded><![CDATA[<p align="justify"><font size="3">Oxytocin (OT) is a neuropeptide that is produced primarily in the hypothalamus and is best known for its role in mammalian birth and lactation. Recent evidence also implicates OT in social behaviors, including parental behavior, the formation of social bonds, and the management of stressful experiences. OT is reactive to stressors, and plays a role in the regulation of both the central and autonomic nervous system, including effects on immune and cardiovascular function. Knowledge of patterns of OT release would be of value in many fields of science and medicine. However, measurements of OT concentration in blood are infrequently performed, and previous attempts to measure OT in saliva have been unsuccessful. Using a sensitive enzyme immunoassay (EIA) and concentrated samples we were able to detect reproducible changes in salivary OT as a function of lactation and massage. These results indicate that measurements of biologically relevant changes in salivary OT are possible. These results confirm the biological relevance of changes in salivary OT with stressors and support saliva as a noninvasive source to monitor central neuroendocrine function.</font></p>

<p align="justify"><font size="3"><strong>Methods</strong></font></p>
<p align="justify"><font size="3">In a double-blind, placebo-controlled, within-subject design, 30 healthy male volunteers were tested for their ability to infer the affective mental state of others using the Reading the Mind in the Eyes Test (RMET) after intranasal administration of 24 IU </font><a href="http://www.oxy-tocin.com/" target="_blank"><font size="3">oxytocin</font></a><font size="3">.<br /></font></p>
<p align="justify"><font size="3"><strong>Results</strong></font></p>
<p align="justify"><a href="http://www.oxy-tocin.com/" target="_blank"><font size="3">Oxytocin</font></a><font size="3"> improved performance on the RMET compared with placebo. This effect was pronounced for difficult compared with easy items.<br /></font></p>

<p align="justify"><font size="3">Although acute treatment with the SSRIs fluoxetine and paroxetine immediately causes increased serotonin levels, delayed ejaculation does not occur. The increased serotonin levels induce </font><a href="http://www.oxy-tocin.com/" target="_blank"><font size="3">oxytocin</font></a><font size="3"> release via activation of 5-HT<sub>1A</sub> receptors, and this might compensate for the inhibitory actions of serotonin on sexual behavior. Chronic treatment with fluoxetine and paroxetine desensitizes 5-HT<sub>1A</sub> receptors on oxytocin neurons, and that might in part determine the onset of delayed ejaculation. Desensitization of 5-HT<sub>1A</sub> receptors is less strong following chronic treatment with the SSRIs fluvoxamine or citalopram, which may attenuate the degree of delayed ejaculation. Conclusions. </font></p>

<title><![CDATA[Absence of oxytocin in the central nervous system of the snake.]]></title>

A high-pressure liquid chromatographic analysis failed to show oxytocin in extracts of hypophysial and brain tissues but provided estimative values of the amounts of vasotocin (12 ng/mg hypophysis and 0.5 ng/mg brain) and mesotocin (500 pg/mg hypophysis and 8 pg/mg brain). Western blots with a polyclonal anti-oxytocin antibody failed to detect oxytocin in both tissues but detected compounds with higher molecular weight than </font><a href="http://www.oxy-tocin.com/" target="_blank"><font size="3">oxytocin</font></a><font size="3">, as well as a relatively weak cross-reactivity with mesotocin. The reverse transcription-polymerase chain reaction analysis failed to detect the expression of oxytocin gene transcript, but detected a transcript related to the mesotocin-neurophysin precursor in both tissues. Immunohistochemistry with the same anti-oxytocin antibody detected strong staining in the neurohypophysis and in few fibers in the inner zone of the median eminence, which was not abolished by pre-adsorption of this antibody with oxytocin, vasopressin, vasotocin or mesotocin and might not be attributed to </font><a href="http://www.oxy-tocin.com/" target="_blank"><font size="3">oxytocin</font></a><font size="3">. In conclusion, our data demonstrate the absence of oxytocin in the central nervous system of the snake <i>B. jararaca</i> and underline the pitfalls that can result from the use of a single technique to investigate the presence of peptides in tissues.</font></p>

We examined the role of serotonin transporter<sup> </sup>(<i>5-HTT</i>) and <a href="http://www.oxy-tocin.com/" target="_blank"><font size="3">oxytocin</font></a><font size="3"> receptor (<i>OXTR</i>) genes in explaining differences<sup> </sup>in sensitive parenting in a community sample of 159 Caucasian,<sup> </sup>middle-class mothers with their 2-year-old toddlers at risk<sup> </sup>for externalizing behavior problems, taking into account maternal<sup> </sup>educational level, maternal </font><a href="http://www.hbcprotocols.com/xrefdepression.html" target="_blank"><font size="3">depression</font></a><font size="3"> and the quality of the<sup> </sup>marital relationship. Independent genetic effects of <i>5-HTTLPR<sup> </sup>SCL6A4</i> and <i>OXTR rs53576</i> on observed maternal sensitivity were<sup> </sup>found. Controlling for differences in maternal education, depression<sup> </sup>and marital discord, parents with the possibly less efficient<sup> </sup>variants of the serotonergic (<i>5-HTT</i> ss) and oxytonergic (AA/AG)<sup> </sup>system genes showed lower levels of sensitive responsiveness<sup> </sup>to their toddlers. Two-way and three-way interactions with marital<sup> </sup>discord or depression were not significant. <title><![CDATA[Oxytocin - a stimulator of directed sperm transport in humans]]></title>

<content:encoded><![CDATA[<p align="justify"><font size="3"><img alt="Male_factor_sperm" hspace="10" src="http://hypericum.files.wordpress.com/2008/05/male-factor-sperm.jpg" align="right" vspace="10" border="0" />Rhythmic peristaltic contractions of the muscular wall of the non-pregnant uterus, as well as rapid sperm transport from the vagina to the Fallopian tubes, have long been documented by means of vaginal sonography and hysterosalpingoscintigraphy. Uterine peristaltic activity reaches a maximum before ovulation and is controlled via oestradiol secretion from the dominant follicle systemically and into the utero-ovarian countercurrent system; it is also enhanced by oxytocin. In this study, the effect of oxytocin and its receptor antagonist atosiban on uterine peristalsis and thus directed sperm transport during the mid and late follicular phases was examined. Atosiban did not show any effect either on frequency or on pattern of the peristaltic contractions. However, oxytocin significantly increased the rapid and directed transport of radiolabelled particles representing spermatozoa from the vagina into the Fallopian tube ipsilateral to the site of the dominant follicle (<i>P</i> = 0.02, 0.04 and 0.02 after 1, 16 and 32 min of documentation respectively). It seems reasonable to assume that oxytocin plays an important, although not critical, role in the mechanisms governing rapid sperm ascension that, at least in humans, were developed to rapidly preserve an aliquot of spermatozoa following intercourse.</font></p>

They have pinpointed&#160;the two brain regions that respond to being betrayed or cheated.</font></p>
<p align="justify"><font size="3">Dr Mauricio Delgado, a psychologist at&#160;Rutgers University in New Jersey said, &#8220;When trust has been broken, something has to allow you to move on with your life and learn to trust again.&#8221; Here something was 'oxytocin. It's this important chemical which helps in maintaining equilibrium between forgiving and forgetting with learning from mistakes." </font></p>
<p align="justify"><font size="3"><img alt="Love_hormone12" hspace="10" src="http://hypericum.files.wordpress.com/2008/05/love-hormone12-small.jpg" align="left" vspace="10"/>In another study Thom&#173;as Baum&#173;gart&#173;ner of the Uni&#173;ver&#173;s&#173;ity of Zu&#173;rich and col&#173;leagues conducted&#160;an study&#160;using 49 male&#160;volunteers who were made to play two types of games --a trust game and a risk game. The volunteers also received brain chemical, oxytocin or a placebo via nasal spray. They were not able to tell the difference between the two. The brains of the volunteers were scanned using functional magnetic resonance imaging (fMRI) during the game. In the first game of trust, the participants were asked to give money, with the understanding that the trustee would invest the money and decide whether to return the profits or betray the subjects' trust and keep all the money. In the second game i.e risk game, the human trustee was replaced by a computer which gave random returns. The researchers said that subjects in first game who received oxytocin were willing to invest again even after their trust was broken by the trustee, while the placebo group became less willing to invest. While in the risk game, the hormone made no difference to the players' investment behavior. "We can see that oxytocin has a very powerful effect," Dr Baumgartner said. "The subjects who received oxytocin demonstrated no change in their trust behavior, even though they were informed that their trust was not honoured in roughly 50% of cases. Our insights into the neural circuitry of trust adaptation and oxytocin's role in trust adaptation, may also contribute to a deeper understanding of mental disorders such as social phobia or autisy that are associated with social deficits. In particular, social phobia (which is the third most common mental health disorder) is characterized by persistent fear and avoidance of social interactions."</font></p>
<p align="justify"><font size="3"><img alt="Love_hormone5" hspace="10" src="http://hypericum.files.wordpress.com/2008/05/love-hormone5-small.jpg" align="right" vspace="10"/>In the experiment, researchers found that oxytocin (OT) decreased activity in two brain regions, only in the trust game and not in risk game. The amygdala was the one region in the brain that processes fear, danger and risk of social betrayal and the other part was striatum, part of the circuitry that guides behaviour based on feedback from rewards. </font></p>
<p align="justify"><font size="3">Researchers showed that the brain hormone, oxytocin affected subjects' responses specifically related to trust. Social phobia or social anxiety disorder is characterized by extreme and persistent anxiety associated with social or performance situations. A person with social phobia experiences anxiety in situations where they are likely to be scrutinised and observed by others. They may have persistent fears about being judged, criticised, ridiculed or humiliated. Scientists believe that the </font><a href="http://www.oxy-tocin.com/" target="_blank"><font color="#0000ff" size="3">amygdala</font></a><font size="3"> is responsible for the symptoms of social Phobia as its the central site in the brain that controls fear responses and produces the symptoms of anxiety disorders. Cognitive-behavior therapy is useful in treating social phobia.</font></p>

Depression may impair social, economical and affective relationships, besides bringing great suffering to the human being. The main symptoms are decreased energy and motivation, anhedonia, sadness, difficulty for concentration and memory, bonding disruptions, etc. The most accepted hypothesis for depression can be well recognized by the main drugs currently adopted for its treatment: they include serotonin and noradrenaline as their fundamental basis. However, their efficacies are still limited. </p>

Female breasts serve both nutritive and sexual functions, unlike other primates they develop at puberty before lactation is necessary. Their sexual attraction is through size, shape and their areolae but also, when unrestrained, their jiggling movements. Small breasts are more sensitive than large. Pregnancy and lactation increase their size. Breast feeding releases oxytocin (milk release) and prolactin (milk secretion), other functions have been proposed for the latter. The pigmented areola contains tubercles of Montgomery that secrete a fluid protecting the skin and creating an olfactory signal for baby and possibly lover; areola corrugation immediately after orgasm physically signals that orgasm has occurred. Male and female (non-pregnant) nipple stimulation has been claimed to induce prolactin secretion but confirmation is needed. Young adult women and men report that breast stimulation not only induces their sexual arousal but enhances it when already aroused.</p>

<title><![CDATA[Oxytocin stimulates colonic motor activity in healthy women]]></title>

The aim of this study was to examine the effect of infusion of oxytocin on colonic motility and sensitivity in healthy women. Fourteen healthy women were investigated twice. A 6-channel perfusion catheter, with three recording points (2 cm apart) proximally and three recording points distally to a barostat balloon, was inserted to the splenic flexure. An intestinal feeding tube was placed in the mid-duodenum. A 90-min duodenal lipid infusion of 3 kcal min<sup>-1</sup> was administered. Thirty minutes after the start of the lipid infusion, the subject randomly received either 20 or 40 mU min<sup>-1</sup> of oxytocin, or isotonic saline as intravenous infusions for 90 min. Meanwhile, the colonic motility was recorded. During the last 30 min of oxytocin and saline infusion, the visceral sensitivity to balloon distensions was examined. During lipid infusion the number of antegrade contractions per hour was 0.7 &#177; 0.3 after saline and 3.9 &#177; 1.4 after oxytocin (<i>P</i> = 0.03), indicating more pronounced lumen-occlusive contractile activity after oxytocin administration. Some of these consisted of high-amplitude (&#62; 103 mmHg in amplitude) antegrade contractions. Lipid infusion evoked a decrease of the balloon volume, reflecting increased colonic tone, but there was no difference between saline and oxytocin. Sensory thresholds did not differ significantly between saline and oxytocin. Infusion of oxytocin stimulates antegrade peristaltic contractions in stimulated colon in healthy women. The effects of oxytocin on colonic motor activity deserve to be further explored, especially in patients with colonic peristaltic dysfunction.</p>

"I had really good highs but some very low lows," Gibson said. "I found out recently I'm manic depressive."</span></p>

McKenzie, who wrote and directed the lively <em>Acting Class Of 1977</em>, which screens on ABC2 on Sunday week, said yesterday that it seemed like a genuine comment but she did not want to delve further. Other NIDA students had no idea that Gibson and Davis would be so successful. "Theatre was very much the focus of our training," McKenzie said. "The thought of people being stars [in film and television] or having stellar careers was not really considered … It was all concentrated on the work and not on what was going to happen down the track."</span></p>
<p align="justify"><span style="font-size:small;">The actress who plays Mystic Marj in the TV series <em>Mortified</em> remembers the young Gibson as "very polite, self-deprecating, shy, certainly talented and just a really nice person … even a little bit embarrassed about the whole acting thing." And Davis, who declined invitations to be in the documentary? McKenzie said she was "intense, private, very talented and idiosyncratic".</span></p>
